Our biggest challenge was to establish procedures and workflows that would allow us to image mitochondria within the calyx in great information using the innovative 3D- EM innovations. After extensive optimization, we designed 2 methods; the first used serial block-face scanning electron microscopy or SBF-SEM for short, which is a type of specialized EM that allows us to create a large-scale set of 3D images in order to examine and rebuild mitochondria within the terminals. 3D reconstructions of images taken with SBF-SEM exposed that mitochondrial volumes within the fully grown calyx and its surrounding axon were considerably higher than those found in its immature equivalent. It also appeared that mitochondria are selectively enhanced within the fully grown calyx, including greater volumes than the surrounding axon. “The strong partnership in between our EM Core and the Young Lab was vital for the success of this work,” notes Naomi Kamasawa, Ph.D., Head of EM Core.
